We are a clinical-stage pharmaceutical company based in Cambridge, Massachusetts. Josh Cohen and Justin Klee co-founded Amylyx in 2013, and we went public on NASDAQ ($AMLX) in January 2022.

Our pipeline includes several therapies under investigation:

  • Avexitide, a GLP-1 receptor antagonist, is ready for Phase 3 and has FDA Breakthrough Therapy designation. It’s in clinical trials for post-bariatric hypoglycemia (PBH), with the Phase 3 LUCIDITY trial starting in April 2025, and for congenital hyperinsulinism (HI).
  • AMX0035 is a combination of sodium phenylbutyrate (PB) and taurursodiol (TURSO). We are investigating this therapy for Wolfram syndrome; positive interim data from the Phase 2 HELIOS trial came out in April 2024, followed by positive topline data in October 2024. AMX0035 has Orphan Drug Designation from both the FDA and the European Commission for Wolfram syndrome. It was previously approved for amyotrophic lateral sclerosis (ALS) in Canada (ALBRIOZA™) in June 2022 and in the U.S. (RELYVRIO®) in September 2022. However, we are voluntarily discontinuing marketing for ALBRIOZA and RELYVRIO as of March-April 2024 after the Phase 3 PHOENIX trial did not meet its primary endpoint. Exploratory analysis from the Phase 2 PEGASUS trial also showed early evidence that AMX0035 affects multiple pathways related to neurodegeneration, including tau, in Alzheimer’s disease.
  • AMX0114 is in development for amyotrophic lateral sclerosis (ALS). The Phase 1 LUMINA clinical trial starts in April 2025. The FDA granted AMX0114 Fast Track designation for ALS in May 2025.
